1. What are Creative Biogene's oligo pools?

Oligo Pool is an innovative, versatile, custom product for genomics discoveries. We synthesize thousands of oligonucleotide sequences at the same time in massive parallel on a microarray chip and then cleave the oligos, releasing them into solution in a single microtube. Synthesis occurs through standard DMT chemistry assuring efficient stepwise yield and high-quality final product. The product is delivered as a pool in a single microtube, ready for use in your experiment.

2. What are some of the applications of oligo pools?

Our customers can then streamline a series of downstream applications within their research with the synthesized oligo pools. These downstream applications include:

  • Generate native DNA sequences
  • Create de novo DNA sequences
  • Build DNA variant libraries
  • Build genes, operons, chassis, pathways and genomes
  • Improve the features of a protein
  • Optimize any antibody via affinity maturation

3. How do you test the quality of the oligo pools?

Each run includes sets of control clusters that are represented across the synthesis chip. These control oligos are pooled, and we NGS verify that the error rate and distribution fall within our QC spec. Customer’s oligo pools are QC'ed by a fragment analysis to ensure the oligos are the correct length, correct concentration and expected mass.

4. What is the error rate for the synthesis of oligos?

Creative Biogene's oligos have an average error rate of around 1:200 or less. Our error rates that come from the control clusters represent the number of base substitutions in the oligos, not the % of full-length oligos etc. We do not routinely see deletions in the synthesis, these are almost certainly only seen after amplification. We can pull error rates for each run post-synthesis, normally about 1/200 bp error.

5. What is unique about your Oligo Pools?

Creative Biogene's oligo pools are with highly accurate synthesis for specific targeting. Our oligo pools demonstrated high uniformity with above 90% of the oligos present within 6x of the mean concentration, which for example in CRISPR applications, ensures highly uniform guide representation. This is important when you are using the pools in reporter assays or CRISPR screens because the quality of the DNA has an influence on downstream processing, screening and analysis.
