Cell Line Description DU4475 is a human breast cancer cell line derived from a cutaneous metastasis in a 70-year-old female patient with advanced breast cancer. The cell line exhibits a lymphoblast-like or rounded morphology and grows primarily in suspension or as loosely adherent cell clusters—characteristics relatively uncommon among breast cancer cell lines. Phenotypically, DU4475 is classified as a triple-negative breast cancer (TNBC) model, as it lacks expression of the estrogen receptor (ER), progesterone receptor (PR), and human epidermal growth factor receptor 2 (HER2). Genetically, the cell line displays an aneuploid karyotype and harbors mutations in key cancer-associated genes, such as BRAF (V600E mutation) and TP53. Due to its unique growth characteristics, TNBC marker profile, and BRAF-mutant genetic background, DU4475 has become a valuable in vitro model for investigating TNBC biology, exploring BRAF signaling dynamics, evaluating kinase-targeted inhibitors, and screening candidate anticancer drugs.

Culture Conditions and Handling
Subculturing 1. Maintain the culture by adding fresh medium or replacing the medium.
2. Establish the culture by centrifuging (1,000 rpm, 5 minutes) and resuspending the cells (at a density of 2 × 10⁵ to 4 × 10⁵ viable cells/mL).
3. Do not allow the cell concentration to exceed 1 × 10⁶ cells/mL.
4. Gently resuspend the cells to prepare a single-cell suspension before subculturing into a new culture vessel.

Subcultivation Ratio Maintain cell density between 2 × 10⁵ and 1 × 10⁶ cells/mL (split ratio approximately 1:2 to 1:4)
