Cell Line Description NCI-H1563 is a human non-small cell lung cancer (NSCLC) cell line derived from a primary lung adenocarcinoma tissue specimen from a male patient. These cells exhibit typical epithelial morphology, growing as an adherent monolayer under standard culture conditions, and retain key biological, cellular, and molecular characteristics of human lung adenocarcinoma. Cytogenetically, NCI-H1563 displays an aneuploid—specifically hypotriploid—karyotype, accompanied by various complex numerical and structural chromosomal alterations. As a widely recognized and valuable in vitro model, this cell line is frequently used to investigate the biological properties of NSCLC, receptor tyrosine kinase signaling, and activated phosphotyrosine pathways involved in tumor growth. Furthermore, NCI-H1563 is extensively utilized in preclinical pharmacological research, including high-throughput screening for novel anticancer drugs, the evaluation of targeted therapies, and functional genomic analysis in lung cancer research.

Culture Conditions and Handling
Subculturing 1. Remove and discard the culture medium.
2. Briefly rinse the cell monolayer with PBS (without Ca2+/Mg2+) to completely remove residual serum.
3. Add 1.0 to 2.0 mL of 0.25% trypsin-0.03% EDTA solution and incubate at room temperature or 37°C until the cell monolayer detaches.
4. Add fresh complete growth medium to neutralize the trypsin and gently pipette up and down to prepare a single-cell suspension.
5. Subculture the cell suspension into new culture vessels.
