Cell Line DescriptionT47D is a human breast cancer cell line established from patients with invasive ductal carcinoma, the most common type of breast cancer. The T47D hypotriploid epithelial cell line was derived from breast ductal carcinoma of a 54-year-old female patient and is a suitable transfection host. The T47D cell line has been used to study a variety of other biological processes, such as hormone signaling, apoptosis, and the effects of environmental toxins on breast cells. The T47D cell line is particularly useful for studying estrogen receptor-positive (ER+) breast cancer because these cells express high levels of estrogen receptors and are responsive to estrogen. The utility of the T47D cell line extends to the formation of xenografts in immunodeficient mice, which is valuable for drug testing, observing changes in receptor status, and studying angiogenesis. Additionally, the T47D cell line is a resource for cancer genetic research, providing insights into the genomic and proteomic landscape that drives breast cancer. By promoting a deeper understanding of the proteomic and transcriptomic profiles of breast cancer, the T47D breast cancer cell line helps identify novel breast cancer cell phenotypes and develop targeted therapies.

Additional InfoT47D cells are unique in the field of cancer research for their hormone expression profile, specifically carrying receptors for 17 beta estradiol, various other steroids, and calcitonin. Furthermore, T47D cells express the WNT7B oncogene. Of note in T47D cells is that, despite abundant intracellular estrogen, their progesterone receptor expression is not modulated by estradiol, which distinguishes them from MCF7 cells.

Culture Conditions and Handling
SubculturingThe volumes used in this protocol are for a 75 cm2 flask. For other sizes of culture vessels, proportionally reduce or increase the amount of dissociation medium.
1. Remove and discard culture medium.
2. Briefly rinse the cell layer with a 0.25% (w/v) trypsin-0.53 mM EDTA solution to remove all traces of serum containing trypsin inhibitors.
3. Add 2.0 to 3.0 mL of trypsin-EDTA solution to the flask and observe the cells under an inverted microscope until the cell layer is dispersed (usually within 5 to 15 minutes). NOTE: To avoid clumping, do not agitate the cells by knocking or shaking the flask while waiting for cells to detach. Cells that are difficult to separate can be placed at 37°C to facilitate dispersion.
4. Add 6.0 to 8.0 mL of complete growth medium and aspirate cells by gentle pipetting.
5. Add appropriate aliquots of cell suspension to new culture vessels.
6. Incubate cultures at 37°C.
