AN3CA Cell Line

General Information
Organism Homo sapiens, human
Cell Line Description AN3CA is a human endometrial adenocarcinoma cell line established in 1962 from a lymph node metastasis of a 55-year-old Caucasian female patient with endometrioid adenocarcinoma of the endometrium associated with acanthosis nigricans. The cells grow as an adherent epithelial monolayer with prominent cytoplasmic vesicles and a doubling time of approximately 45 to 55 hours. AN3CA is characterized by a homozygous PTEN truncating mutation and is microsatellite instability-high owing to defective mismatch repair, making it a standard model for PTEN-deficient endometrial cancer. Additional alterations include TP53, PIK3R1 and MAPK3 mutations. The line is negative for estrogen and progesterone receptors, is tumorigenic in nude mice and in the hamster cheek pouch, and is widely used in PARP inhibitor sensitivity studies, PI3K/AKT/mTOR pathway inhibitor screening, and endometrial cancer xenograft modeling. It remains one of the few widely available endometrial carcinoma lines carrying biallelic PTEN loss, and is used to model type I endometrioid disease with defective mismatch repair and to evaluate DNA damage response inhibitors.

Applications 1. Modeling of PTEN-deficient endometrial cancer and investigation of sensitivity to PARP inhibitors;
2. Screening of inhibitors targeting the PI3K/AKT/mTOR pathway (including rapamycin and dual mTORC1/2 inhibitors);
3. Development of xenograft models for TP53-mutant and non-endometrioid endometrial cancers;
4. Evaluation of combination therapies targeting the Ras/MAPK and PI3K pathways;
5. Biological characteristics of hormone receptor-negative endometrial cancer.

Culture Conditions and Handling
Subculturing 1. Remove and discard the culture medium.
2. Briefly rinse the cell monolayer with PBS (Ca²⁺/Mg²⁺-free) to thoroughly remove residual serum.
3. Add 2.0 to 3.0 mL of 0.25% Trypsin–0.53 mM EDTA solution, and observe under an inverted microscope until the cell monolayer has dispersed (typically takes 2 to 5 minutes).
4. Add 6.0 to 8.0 mL of complete growth medium, and gently pipette to prepare a single-cell suspension.
5. Aliquot the cell suspension into new culture vessels.
Medium Renewal Every 3 to 4 days
Subcultivation Ratio Split confluent cultures at a ratio of 1:3 to 1:8 once or twice weekly; seed at 1–2 × 10⁶ cells per 25 cm².
Culture Conditions Atmosphere: Air, 95%; CO2, 5%; Temperature: 37°C
Cryopreservation 70% complete growth medium + 20% FBS + 10% DMSO
