MOLT-4 Cell Line

Cell Line Description MOLT-4 is an established human T-lymphoblastic cell line, originally generated by J. Minowada and colleagues in 1971. The cell line was isolated from the peripheral blood of a 19-year-old Caucasian male patient experiencing a relapse of acute lymphoblastic leukemia (ALL) following multi-drug combination chemotherapy. MOLT-4 cells exhibit a round, lymphoid morphology and grow exclusively in suspension, appearing either as single cells or as irregular clusters. The cell line is globally recognized as a fundamental and iconic in vitro model for studying T-cell leukemia and the pathogenesis of human immunodeficiency virus (HIV-1). Furthermore, MOLT-4 is a key member of the renowned NCI-60 cancer cell line panel, serving as a cornerstone in global high-throughput anti-tumor drug development and the identification of molecular targets.

Characteristics
Tumorigenic Yes, forms tumors in untreated nude mice, anti-lymphocyte serum-treated mice, and X-irradiated mice.
Karyotype Human flat-moded hypertetraploid karyotype; modal chromosome count typically shifts around 95 (ranges between 89–99).
Immunophenotype T-cell markers (cyCD3, CD2, CD3, CD4, CD5, CD6, CD7, and CD8) were strongly positive; the myeloid marker CD13, the B-cell marker CD19, and TCRα/β or TCRγ/δ complexes all tested negative; and terminal deoxynucleotidyl transferase (TdT) showed high levels of endogenous expression.
Growth Kinetics Rapid suspension expansion; typical population doubling time spans approximately 40 hours. Viability may transiently drop immediately after thawing recovery.

Culture Conditions and Handling
Subculturing 1. MOLT-4 cells grow in loose suspension aggregates; enzymatic dissociation with trypsin is not required.
2. Gently agitate the culture vessel to ensure a homogeneous cell suspension, then transfer the medium to a sterile conical centrifuge tube.
3. Centrifuge at approximately 200-300 × g for 5 minutes.
4. Completely aspirate the supernatant, retaining the compact cell pellet.
5. Resuspend the cells evenly in pre-warmed, fresh complete medium.
6. Transfer to a new culture vessel.
Medium Renewal Every 2 to 3 days depending on metabolic rate and acid pH visual indicators.
Subcultivation Ratio Split optimal suspension cultures at a ratio of 1:2 to 1:3.
Culture Conditions Atmosphere: Air, 95%; CO2, 5%; Temperature: 37°C
Cryopreservation 90% Complete growth medium + 10% DMSO (or 50% RPMI 1640 + 40% FBS + 10% DMSO)
