Cell Line DescriptionEstablished in 1970 from metastatic cells found in the bone marrow aspirate of a four-year-old female, the SK-N-SH cell line is a well-known human neuroblastoma cell line that is widely used in biomedical research. Treatment with all-trans retinoic acid causes these cells to differentiate and adopt a neuronal phenotype characterized by extensive neurite outgrowth. This makes them particularly suitable for delineating signaling pathways involved in neuronal differentiation. In addition, SK-N-SH cells are known to form tumors in immunocompromised mice. SK-N-SH cells have played a crucial role in elucidating the molecular mechanisms behind various neurological diseases and conditions, including Alzheimer's disease, Parkinson's disease, and neurodevelopmental disorders. Known for their ability to differentiate into mature neuron-like cells, they are a valuable tool in drug discovery and screening assays aimed at identifying potential treatments for neurological diseases. Additional InfoSK-N-SH cells are characterized by high dopamine-β-hydroxylase activity. Very low levels of activity for glutamate decarboxylase (the enzyme responsible for converting glutamate to GABA) were also found in this cell line. Choline acetyltransferase activity was negligible in this cell line. These findings indicate that SK-N-SH cells exhibit a neuronal phenotype and possess multiple neurochemical markers.

Culture Conditions and Handling
Subculturing1. Remove old medium from adhered cells and wash with PBS without calcium and magnesium. For T25 flasks, use 3-5 ml PBS and for T75 flasks, use 5-10 ml.
2. Then completely cover the cells with Accutase, using 1-2 ml for T25 flasks and 2.5 ml for T75 flasks.
3. Let the cells incubate at room temperature for 8-10 minutes to allow them to detach.
4. After incubation, gently mix the cells with 10 ml medium to resuspend, then centrifuge at 300 x g for 3 minutes.
5. Discard the supernatant, resuspend the cells with fresh medium, and transfer them to a new flask already containing fresh medium.

Subcultivation RatioThe ratio of 1:2 to 1:4 is recommended.
Culture MediumEMEM (EBSS) + 2 mM Glutamine + 10% Foetal Bovine Serum (FBS).
