HCC38 Cell Line

General Information
Organism Homo sapiens, human
Cell Line Description HCC38 is a human ductal breast carcinoma cell line derived from the primary breast tumor of a 50-year-old female patient. These cells exhibit an epithelial morphology and grow as an adherent monolayer under standard culture conditions. Phenotypically, HCC38 is classified as a model of basal-like triple-negative breast cancer (TNBC), characterized by the absence of estrogen receptor (ER) and progesterone receptor (PR) expression and a lack of human epidermal growth factor receptor 2 (HER2) overexpression. Cytogenetically, the cell line displays an aneuploid karyotype with complex numerical and structural chromosomal alterations. At the molecular level, HCC38 harbors a mutation in the TP53 tumor suppressor gene. With its well-defined triple-negative phenotype and unique genomic profile, HCC38 serves as a valuable in vitro model for investigating the pathogenesis of TNBC, exploring signaling dynamics in the basal-like subtype, evaluating targeted therapies and candidate chemotherapeutic agents, and conducting high-throughput functional genomic screens.

Culture Conditions and Handling
Subculturing 1. Remove and discard the culture medium.
2. Briefly rinse the cell monolayer with PBS (without Ca2+/Mg2+) to completely remove residual serum.
3. Add 1.0 to 2.0 mL of 0.25% trypsin-0.03% EDTA solution and incubate at 37°C until the cell monolayer detaches (typically 2 to 5 minutes).
4. Add fresh complete growth medium to neutralize the trypsin and gently pipette up and down to prepare a single-cell suspension.
5. Subculture the cell suspension into new culture vessels.
Medium Renewal Every 2 to 3 days
Subcultivation Ratio A split ratio of 1:3 to 1:6 is recommended
Culture Conditions Atmosphere: Air, 95%; CO2, 5%; Temperature: 37°C
Cryopreservation Complete growth medium supplemented with 5% to 10% (v/v) DMSO
