Cell Line DescriptionSW480 is a human colon adenocarcinoma cell line established from primary tumors of colorectal cancer patients. The SW480 epithelial cell line was isolated from a colon tumor from a 50-year-old white male patient with colorectal adenocarcinoma. The SW-480 cell line is sensitive to human immunodeficiency virus 1 and expresses high levels of p53 protein. In addition, SW480 cells positively express c-myc, K-ras, H-ras, N-ras, myb, sis, and fos oncogenes. The SW480 cell line is tumorigenic in nude mice and produces carcinoembryonic antigen (CEA) and keratin. The SW480 cell line has been widely used as a model system to study the biology of colorectal cancer and develop new treatments for the disease. Researchers use these cells to study the molecular mechanisms of tumor growth, invasion and metastasis, as well as drug resistance and potential therapeutic targets. The use of tumor cells injected into mice has been used for decades as a model for evaluating preclinical treatments with xenograft tumor systems.

Additional InfoSW-480 cells have an epithelial-like morphology, and their xenografts form gland-like structures. Furthermore, consistent with being at an early stage of tumor progression, SW480 is less metastatic, less tumorigenic, and exhibits less resistance to FAS- or TNF-α-induced apoptosis. The SW480 xenograft model is a preclinical model used in studies examining therapeutics targeting the β-catenin pathway, such as silibinin. The SW480 model is also suitable for studying the association between human epidermal growth factor receptor (EGFR) expression levels and anti-tumor growth therapies such as cetuximab.

Culture Conditions and Handling
SubculturingThe volumes used in this protocol are for a 75 cm2 flask. For other sizes of culture vessels, proportionally reduce or increase the amount of dissociation medium.
1. Remove and discard culture medium.
2. Briefly rinse the cell layer with a 0.25% (w/v) trypsin-0.53 mM EDTA solution to remove all traces of serum containing trypsin inhibitors.
3. Add 2.0 to 3.0 mL of trypsin-EDTA solution to the flask and observe the cells under an inverted microscope until the cell layer is dispersed (usually within 5 to 15 minutes). NOTE: To avoid clumping, do not agitate the cells by knocking or shaking the flask while waiting for cells to detach. Cells that are difficult to separate can be placed at 37°C to facilitate dispersion.
4. Add 6.0 to 8.0 mL of complete growth medium and aspirate cells by gentle pipetting.
5. Add appropriate aliquots of cell suspension to new culture vessels.
6. Incubate cultures at 37°C. No carbon dioxide.
