Cell Line DescriptionRS4-11 is a lymphoblastoid cell line isolated from the bone marrow of a 32-year-old Caucasian female patient with acute lymphoblastic leukemia. It is useful in the study of immunology and immune system disorders. These cells lack surface and cytoplasmic immunoglobulins and are negative for CALLA (CD10). RS4-11 cells are also negative for myeloperoxidase and chloroacetate esterase and do not stain with Sudan black. In addition, these cells are strongly positive for terminal deoxynucleotidyl transferase (TdT) and are positive for BA-1 (CD24), BA-2 (CD9), and MHC class II antigens (HLADR).

Additional InfoThe RS4-11 cell line is a suitable in vitro model for studying leukemias carrying t(4;11) because the original cytogenetic abnormality is present and preserved. As with other cell lines, the karyotype evolves during long-term culture through the accumulation of secondary mutations and rearrangements.

Culture Conditions and Handling
SubculturingCultures can be maintained by adding fresh medium or changing the medium. Alternatively, cultures can be established by centrifugation and resuspending at 1 X 10^5 viable cells/mL. Maintain cell density between 1 X 10^5 and 1 X 10^6 viable cells/mL.
Medium RenewalEvery 2 to 3 days.
Subcultivation RatioThe ratio of 1:2 to 1:4 is recommended.
