REC1 Cell Line

Cell Line Description REC1 is a human mantle cell lymphoma (MCL) cell line derived from the peripheral blood of a 61-year-old male patient with aggressive MCL. REC1 cells exhibit a lymphoblast-like morphology and grow primarily in suspension or as loose cell aggregates. The cell line harbors the hallmark chromosomal translocation t(11;14)(q13;q32), which juxtaposes the CCND1 gene with the immunoglobulin heavy chain locus, thereby driving the characteristic overexpression of cyclin D1. Additionally, REC1 carries mutations in key cancer-driver genes, including TP53 and ATM, reflecting the high genomic instability typical of refractory MCL. With its well-defined cytogenetic profile and stable growth characteristics, REC1 has become a valuable in vitro model for investigating MCL pathogenesis, evaluating novel targeted therapies (such as BTK inhibitors), exploring mechanisms of drug resistance, and conducting functional genomic screens.

Culture Conditions and Handling
Subculturing 1. Maintain the culture by adding fresh medium or replacing the medium.
2. Establish the culture by centrifuging (1,000 rpm, 5 minutes) and resuspending the cells (viable cell density of 3 × 10⁵ to 5 × 10⁵ cells/mL).
3. Do not allow the cell concentration to exceed 1.5 × 10⁶ cells/mL.
4. Gently resuspend the cells to prepare a single-cell suspension before subculturing into a new culture vessel.

Subcultivation Ratio Maintain cell density between 3 × 10⁵ and 1.5 × 10⁶ cells/mL (split ratio approximately 1:2 to 1:4)
