Cell Line DescriptionNCI-H82 is an epithelioid cell line isolated from the lung of a 40-year-old Caucasian male with lung cancer. The cell line was isolated in 1978 by A.F. Gazdar and colleagues from the pleural effusion of a patient with small cell lung cancer. The morphology of the original tumor does not conform to the characteristics of small cell lung cancer (SCLC). The cell line is biochemically and morphologically a variant of SCLC, expressing neuron-specific enolase and the brain isoenzyme of creatine kinase. It has no detectable levels of L-DOPA decarboxylase or bombardin. NCI-H82 cells produce an abnormally sized p53 mRNA (3.7 kb). The c-myc DNA sequence is amplified approximately 25-fold and c-myc RNA is increased 24-fold compared to normal cells. NCI-H82 cells are reported to express functional ANP receptors, but treatment with ANP does not alter their growth pattern.

Addtional InfoThe NCI-H82 cells stain positively for neurofilaments and vimentin. There is expression of v-fes, v-fms, Ha-ras, Ki-ras, N-ras and c-raf 1 mRNAs.
Characteristics
KaryotypeThis is a near-triploid human cell line. The modal chromosome number is 58, the prevalence is 44%, and the prevalence of polyploidy is 3%. Each cell has two copies of the normal X chromosome. No Y chromosome was detected in the Q-banded preparation.

Culture Conditions and Handling
SubculturingMaintain the culture by regularly adding or replacing the medium. Start the culture at 2 x 10^5 cells/mL and maintain the cell concentration in the range of 1 x 10^5 to 1 x 10^6 cells/mL for optimal growth.
Medium Renewal2 to 3 times per week
Subcultivation RatioThe ratio of 1:3 to 1:5 is recommended.
Culture MediumRPMI 1640, w: 2.1 mM stable Glutamine, w: 2.0 g/L NaHCO3. To make the complete growth medium, add the following components to the base medium: fetal bovine serum to a final concentration of 10%.
