Cell Line Description KTC-1 is a human papillary thyroid carcinoma (PTC) cell line established in 1991, derived from the pleural effusion of a 68-year-old male patient with recurrent metastatic PTC. As a well-characterized model, it is widely utilized to investigate the progression and underlying molecular mechanisms of thyroid malignancies. KTC-1 cells exhibit an epithelial morphology and are known to harbor the BRAF V600E mutation, which leads to the constitutive activation of the MAP kinase signaling pathway. This cell line is particularly valuable in the following research areas: exploring the biological behavior of advanced PTC, evaluating redifferentiation therapies (specifically NIS induction), and screening for novel kinase inhibitors or radiosensitizers targeting metastatic disease.

Culture Conditions and Handling
Subculturing 1. Remove and discard the culture medium.
2. Briefly rinse the cell monolayer with PBS (Ca²⁺/Mg²⁺-free) to thoroughly remove residual serum.
3. Add 2.0 to 3.0 mL of 0.25% Trypsin–0.53 mM EDTA solution, and observe under an inverted microscope until the cell monolayer has dispersed (typically takes 5 to 10 minutes).
4. Add 6.0 to 8.0 mL of complete growth medium and gently pipette to prepare a single-cell suspension.
5. Aliquot the cell suspension into new culture vessels.
