DNA methylation is also a usual subject of agrigenomic investigations into responses to temperature extremes, drought, and other environmental changes.

We can offer the following four types of methylation analyses which are based on the treatment of genomic DNA with sodium bisulfite, including Methylation Specific PCR, Bisulfite Sequencing, Bisulfite Pyrosequencing and Genome Wide Methylation Analysis.

3. What is the principle of methylation analyses?

When treat genomic DNA with sodium bisulfite, the process deaminates unmethylated cytosine residues to uracil leaving methylated cytosine residues unchanged. After PCR, uracil hydrogen bonds to adenine that will then hydrogen bond to thymine. Thus, the concept is that unmethylated cytosines will become thymines and methylated cytosines will still remain cytosines in the genomics sequence.

4. What are our DNA methylation analysis methods?

1) Methylation sequencing with next generation sequencing (NGS). NGS enables comprehensive profiling of methylation patterns at single-base resolution throughout the whole genome, or in targeted epigenetic regions of interest.

2) Methylation microarrays. Arrays enable quantitative interrogation of selected methylation sites throughout the genome, providing high-throughput capabilities that minimize the cost per sample.

Pyrosequencing facilitates accurate DNA methylation analysis through:

1) Measuring methylation status of individual and multiple CpG or non-CpG (CpN) sites

2) Quantifying methylation in explicit sequence context

4) Permitting rapid and high-throughput analysis of methylation
