NCI-H1944 Cell Line

General Information
Organism Homo sapiens, human
Cell Line Description NCI-H1944 is a human non-small cell lung cancer (adenocarcinoma subtype) cell line, established in June 1988 at the NCI–Navy Medical Oncology Branch—a joint venture between the National Cancer Institute (NCI) and the U.S. Navy. The cell line was derived from a soft-tissue metastasis in a 62-year-old Caucasian female patient who had stage 3B disease and a prior history of radiation therapy. These cells grow as an adherent monolayer and exhibit epithelial-like characteristics. Their genomic profile includes a heterozygous KRAS p.Gly13Asp activating mutation, a truncating ATR variant, and additional alterations in the STK11, KEAP1, CDKN2A, and RBM10 genes. The cell line is microsatellite-stable (MSS), near-diploid, and capable of forming subcutaneous tumors in athymic nude mice. NCI-H1944 is widely used in research areas such as signaling pathways in KRAS-mutant lung cancer, KRAS-targeted therapeutic strategies, receptor tyrosine kinase (RTK) profiling, three-dimensional (3D) spheroid culture, and high-throughput pharmacogenomic screening. Furthermore, it is utilized in studies involving the co-targeting of MYC and HIF-1α, as well as in the preclinical in vivo evaluation of candidate therapeutic agents for KRAS-mutant lung cancer, frequently serving as a reference model within research panels for this cancer type.

Culture Conditions and Handling
Subculturing 1. Remove and discard the culture medium.
2. Briefly rinse the cell monolayer with PBS (Ca²⁺/Mg²⁺-free) to thoroughly remove residual serum.
3. Add 2.0 to 3.0 mL of 0.25% Trypsin–0.53 mM EDTA solution, and observe under an inverted microscope until the cell monolayer has dispersed (typically takes 2 to 5 minutes).
4. Add 6.0 to 8.0 mL of complete growth medium, and gently pipette to prepare a single-cell suspension.
5. Aliquot the cell suspension into new culture vessels.
Medium Renewal Every 2 to 3 days
Subcultivation Ratio A split ratio of 1:3 to 1:6 is recommended.
Culture Conditions Atmosphere: Air, 95%; CO2, 5%; Temperature: 37°C
Cryopreservation Complete growth medium supplemented with 5% (v/v) DMSO
