OC 316 Cell Line

General Information
Organism Homo sapiens, human
Cell Line Description OC 316 is a human ovarian cancer cell line derived from the malignant ascites of a female patient diagnosed with advanced-stage ovarian serous adenocarcinoma. OC 316 cells exhibit an epithelial-like morphology and grow as an adherent monolayer under standard culture conditions. The cell line retains key biological and phenotypic characteristics of high-grade ovarian serous carcinoma, making it a valuable model for gynecologic oncology research. Cytogenetically, OC 316 displays an aneuploid karyotype characterized by complex numerical and structural chromosomal rearrangements. At the molecular level, the cell line harbors genetic alterations in key tumor suppressor genes and oncogenic pathways (such as TP53 mutations) commonly implicated in the pathogenesis of ovarian cancer. Given its well-defined malignant origin and reliable in vitro growth characteristics, OC 316 has become an important research model for investigating ovarian cancer biology, studying mechanisms of platinum resistance, evaluating targeted therapies, and conducting functional genomic screens.

Culture Conditions and Handling
Subculturing 1. Remove and discard the culture medium.
2. Briefly rinse the cell monolayer with PBS (without Ca2+/Mg2+) to completely remove residual serum.
3. Add 1.0 to 2.0 mL of 0.25% trypsin-0.03% EDTA solution and incubate at 37°C until the cell monolayer detaches (typically 2 to 5 minutes).
4. Add fresh complete growth medium to neutralize the trypsin and gently pipette up and down to prepare a single-cell suspension.
5. Subculture the cell suspension into new culture vessels.
Medium Renewal Every 2 to 3 days
Subcultivation Ratio A split ratio of 1:3 to 1:6 is recommended
Culture Conditions Atmosphere: Air, 95%; CO2, 5%; Temperature: 37°C
Cryopreservation Complete growth medium supplemented with 5% to 10% (v/v) DMSO
