| Cell Line Description |
OCI-LY3 is a human diffuse large B-cell lymphoma (DLBCL) cell line belonging to the activated B-cell-like (ABC) subtype. Established in 1983 at the Ontario Cancer Institute, the cell line was derived from the bone marrow of a 52-year-old Caucasian male patient who was experiencing a disease relapse (stage IVB). In suspension culture, these cells grow as single, round cells or in small clusters. OCI-LY3 has become a reference model for studying persistent B-cell receptor (BCR) signaling in ABC-DLBCL. The cell line exhibits an immunophenotype characteristic of mature B cells, expressing surface IgG lambda and cytoplasmic IgM lambda; it also expresses CD19, CD20, CD37, CD38, CD80, and HLA-DR, while lacking expression of CD3, CD10, CD34, and CD138. OCI-LY3 is widely used in research concerning the dysregulation of the NF-κB pathway in ABC-DLBCL, responses to BTK and BCL2 inhibitors, and mechanisms of chemotherapy resistance. Furthermore, it serves as a key cell line in large-scale pharmacogenomic screens and CRISPR-based functional genomic screens. |

| Culture Conditions and Handling |
| Subculturing |
1. Collect the cell suspension into a centrifuge tube and centrifuge at approximately 200 × g for 5 minutes. 2. Carefully discard the supernatant. 3. Resuspend the cell pellet in fresh complete growth medium and gently pipette to break up any cell aggregates. 4. Count the cells and adjust to the recommended seeding density with fresh medium. 5. Aliquot the cell suspension into new culture vessels. |
| Medium Renewal |
2 to 3 times per week |
| Subcultivation Ratio |
Maintain cells at 0.1-0.8 × 10⁶ cells/mL; split at a ratio of 1:3 to 1:5 every 2 to 3 days. |
| Culture Conditions |
Atmosphere: Air, 95%; CO2, 5%; Temperature: 37°C |
| Cryopreservation |
Use a defined serum-free cryopreservation medium, or 70% complete growth medium + 20% FBS + 10% DMSO |
