Cell Line Description HCCLM3 is a highly metastatic human hepatocellular carcinoma (HCC) cell line. It was established using the MHCC97-H cell line as the parent cell line through serial lung metastasis in nude mice. HCCLM3 is characterized by its extremely high spontaneous metastasis rate, particularly to the lungs, making it an important model for studying the molecular mechanisms of cancer invasion, epithelial-mesenchymal transition (EMT), and metastasis. It is widely used to evaluate the efficacy of anti-angiogenic drugs and targeted therapies in advanced liver cancer.

Culture Conditions and Handling
Subculturing 1. Remove and discard the culture medium.
2. Quickly wash the cell layer with Ca2+/Mg2+-free PBS buffer to remove residual serum.
3. Add 1.0 to 2.0 mL of 0.25% trypsin-EDTA solution and incubate at 37°C until cells detach (usually 3 to 5 minutes).
4. Neutralize with complete culture medium and gently pipette to obtain a single-cell suspension.
5. Aliquot the cells into new culture dishes.
Medium Renewal 2 to 3 times per week
Subcultivation Ratio A split ratio of 1:3 to 1:6 is recommended
Culture Medium DMEM (High Glucose) supplemented with 10% Fetal Bovine Serum (FBS) and 1% Penicillin-Streptomycin.
Culture Conditions Atmosphere: Air, 95%; CO2, 5%; Temperature: 37°C
Cryopreservation Complete growth medium supplemented with 5% to 10% (v/v) DMSO
