Cell Line Description bEnd.3 is a mouse brain capillary endothelial cell line derived from T antigen-converted polyomaviruses in BALB/c mice. Because it maintains many morphological and functional characteristics of primary brain endothelial cells, it is currently the most widely used in vitro model of the blood-brain barrier (BBB). bEnd.3 cells can form a tightly connected fused monolayer and express BBB-specific transport proteins and receptors. This cell line is an important tool for studying cerebral vascular permeability, leukocyte adhesion, and the transport of drugs and nutrients across the BBB.

Culture Conditions and Handling
Subculturing 1. Remove and discard the culture medium.
2. Briefly rinse the cell layer with PBS (without Ca2+/Mg2+) to remove all traces of serum.
3. Add 2.0 to 3.0 mL of 0.25% Trypsin - 0.53 mM EDTA solution and observe under an inverted microscope until the cell layer is dispersed (usually 3 to 8 minutes).
4. Add 6.0 to 8.0 mL of complete growth medium and gently pipette to obtain a single-cell suspension.
5. Aliquot cell suspension into new culture vessels.
Medium Renewal 2 to 3 times per week
Subcultivation Ratio A split ratio of 1:3 to 1:10 is recommended
Culture Medium Dulbecco's Modified Eagle's Medium (DMEM) with high glucose (4.5 g/L), supplemented with 10% Fetal Bovine Serum (FBS).
Culture Conditions Atmosphere: Air, 95%; CO2, 5%; Temperature: 37°C
Cryopreservation Complete growth medium supplemented with 5% to 10% (v/v) DMSO
