Cell Line DescriptionThe NALM6 cell line was derived from cultures of the blood of a 19-year-old male patient diagnosed with non-T, non-B acute lymphoblastic leukemia (ALL) during relapse. These cultures generated eight "leukemic" cell lines and two "normal B" cell lines. Interestingly, when cultured, the leukemic blasts displayed a unique membrane phenotype characterized by the presence of SmIg (lambda and mu chain immunoglobulins), Ia-like B-cell-associated antigen (Ia), and common forms of ALL-specific antigens (cALL). The Nalm6 cell line is a reliable model to study the complex nature of acute leukemia. It has facilitated the study of gene expression profiles involved in glycolysis, lipid and carbohydrate metabolism, and the mTORC1 pathway, highlighting metabolic reprogramming in leukemic cells. Furthermore, the use of this cell line in reverse genetics and whole transcriptome analyses has helped to dissect the complex molecular networks driving leukemia progression and drug resistance.

Additional InfoThe sensitivity of the NALM6 cell line to cytotoxic agents and its role in elucidating the function of homologous recombination (HDR) repair is of particular interest, especially with regard to the ability of HDR cells to correct DNA damage. Studies utilizing the NALM6 cell line, including studies of clonal variants such as clone G5 and drug-resistant cell lines such as C9 with a high frequency of HPRT mutations or with a resistance index, have provided insights into leukemia heterogeneity.
