Cell Line Description SK-MEL-1 is a human malignant melanoma cell line isolated in 1968 from the thoracic duct lymph nodes of a 29-year-old Caucasian male with extensive metastatic melanoma. Unlike many other melanoma cell lines that grow in an adherent monolayer, SK-MEL-1 is characterized by suspension growth, often forming large, spherical cell clusters. This cell line is renowned for its high melanin-producing capacity and is frequently used to study melanin production, tumor antigen expression, and the efficacy of immunotherapies. It is an important model for studying the biology of metastatic melanoma and developing targeted therapies for advanced skin cancer.

Culture Conditions and Handling
Subculturing 1. Cell culture can be maintained by adding fresh culture medium or changing the culture medium.
2. Alternatively, cell culture can be established by centrifugation and resuspending in fresh culture medium.
3. To maintain optimal growth, the cell density should be maintained between 2 × 10⁵ and 1 × 10⁶ viable cells/mL.
4. The cell density should not exceed 1 × 10⁶ cells/mL.
Medium Renewal Every 2 to 3 days
Subcultivation Ratio A split ratio of 1:2 to 1:4 is recommended
Culture Medium Eagle's Minimum Essential Medium (EMEM) or RPMI 1640 supplemented with 10% to 15% Fetal Bovine Serum (FBS).
