Cell Line Description The J82 cell line, derived from a 58-year-old Caucasian male patient with high-grade (T2 stage) transitional cell carcinoma of the bladder, is one of the most widely used models for studying the biology of bladder cancer. This cell line is characterized by its ability to grow as an adherent monolayer with highly rearranged karyotypes. J82 cells are known to express a variety of tumor-associated antigens and are frequently used to study the mechanisms of bladder cancer progression, chemotherapy resistance, and the development of targeted therapies.

Culture Conditions and Handling
Subculturing 1. Remove and discard the culture medium.
2. Briefly rinse the cell layer with Ca²⁺/Mg²⁺-free PBS or 0.25% trypsin-EDTA solution to remove residual serum.
3. Add 2.0 to 3.0 mL of trypsin-EDTA solution and observe the cells under a microscope until the cell layer disperses (5 to 15 minutes).
4. Add 6.0 to 8.0 mL of complete culture medium and gently aspirate the cells using a pipette.
5. Aliquot the cell suspension into new culture dishes.

Subcultivation Ratio A split ratio of 1:2 to 1:4 is recommended
Culture Medium Eagle's Minimum Essential Medium (EMEM) supplemented with 10% Fetal Bovine Serum (FBS)
