HCA-7 Cell Line

General Information
Organism Homo sapiens, human
Cell Line Description HCA-7 is a human colorectal adenocarcinoma cell line established in 1985 by Susan C. Kirkland in the Department of Histopathology at Imperial College London. The cell line was derived from a primary colon adenocarcinoma in a 58-year-old female patient; the tumor was moderately differentiated and classified as Dukes stage B. These cells grow as an adherent, polarized epithelial monolayer, characterized by the presence of apical tight junctions and desmosomes. Due to directional fluid transport, they can spontaneously form "hemicysts" or dome-like structures. HCA-7 cells constitutively express cyclooxygenase-2 (COX-2) and secrete prostaglandin E2 (PGE2) and other prostaglandins into the basolateral space, making them an important model for studying the biology of COX-2 and prostaglandins in colorectal cancer. The cell line exhibits microsatellite instability resulting from heavy methylation of the MLH1 promoter. It carries truncating mutations in the APC and FBXW7 genes, while the KRAS, BRAF, PIK3CA, and EGFR genes remain wild-type. When injected subcutaneously into nude mice, the cells form adenocarcinomas. A clonal derivative, "HCA-7 Colony 29," is capable of forming a uniform, polarized monolayer on permeable supports, making it the preferred model for directional transport assays; both the parental line and this clone retain the COX-2 phenotype.

Characteristics
Tumorigenic Yes, forms adenocarcinomas in nude mice following subcutaneous inoculation
Karyotype Chromosome numbers 41–82 with a modal number of 43; approximately 75% of cells fall in the 43–46 range
Expression Markers Constitutively expresses COX-2 (PTGS2); releases prostaglandin E2, PGD2, TXA2 and PGF2α basolaterally; expresses EGFR and amphiregulin; secretes CEA at approximately 91–100 ng per mL per 24 h from a confluent 25 cm² monolayer. MLH1 protein is not detectable by immunohistochemistry
Culture Conditions and Handling
Subculturing 1. Remove and discard the culture medium.
2. Briefly rinse the cell monolayer with PBS (Ca²⁺/Mg²⁺-free) to thoroughly remove residual serum.
3. Add 2.0 to 3.0 mL of 0.25% Trypsin–0.53 mM EDTA solution, and observe under an inverted microscope until the cell monolayer has dispersed (typically takes 2 to 5 minutes).
4. Add 6.0 to 8.0 mL of complete growth medium, and gently pipette to prepare a single-cell suspension.
5. Aliquot the cell suspension into new culture vessels.
Medium Renewal 2 times per week
Subcultivation Ratio Split sub-confluent (70–80%) cultures at a ratio of 1:2 to 1:10 using 0.05% trypsin/EDTA; seed at 2–4 × 10⁴ cells/cm².
Culture Conditions Atmosphere: Air, 92.5%; CO2, 7.5%; Temperature: 37°C
Cryopreservation Complete growth medium supplemented with 5% (v/v) DMSO
