Cell Line Description SMMC-7721 is a human hepatocellular carcinoma (HCC) cell line established in 1977, derived from a 50-year-old male patient with primary liver cancer. The human liver cancer cell line SMMC-7721 expresses both EGFR and VEGFR, making it an indispensable tool in liver cancer research. These cells exhibit an epithelial-like morphology, characterized by rapid growth rates and stable biological properties under in vitro culture conditions. SMMC-7721 is frequently utilized to investigate the molecular mechanisms of hepatocarcinogenesis and tumor cell metabolism, as well as to evaluate the efficacy of novel anticancer agents—including traditional Chinese medicine extracts and targeted small-molecule drugs. It has become a standard model for studying the progression of hepatic malignancies and therapeutic interventions.

Culture Conditions and Handling
Subculturing 1. Remove and discard the culture medium.
2. Briefly rinse the cell layer with PBS (without Ca2+/Mg2+) to remove all traces of serum.
3. Add 2.0 to 3.0 mL of 0.25% Trypsin - 0.53 mM EDTA solution and observe under an inverted microscope until the cell layer is dispersed (usually 3 to 8 minutes).
4. Add 6.0 to 8.0 mL of complete growth medium and gently pipette to obtain a single-cell suspension.
5. Aliquot cell suspension into new culture vessels.

Subcultivation Ratio A split ratio of 1:3 to 1:8 is recommended
