| Cell Line Description |
AML-12 (Alpha Mouse Liver 12) is a non-malignant, immortalized mouse hepatocyte cell line established in 1994. This cell line was derived from the liver of a CD-1 (ICR) transgenic mouse carrying a human transforming growth factor-alpha (TGF-α) transgene driven by the mouse albumin promoter (Tg(Alb-1TGFa)22Bri). AML-12 cells grow as an adherent monolayer and exhibit the typical polygonal, epithelial-like morphology of hepatocytes. In the fields of hepatology, toxicology, and metabolic research, AML-12 is recognized as a high-quality, "gold standard" model of non-tumorigenic, functional mouse hepatocytes. Unlike hepatocellular carcinoma cell lines, AML-12 cells stably maintain differentiated hepatocyte functions, including serum protein synthesis, peroxisome proliferation, active glycogen storage, and inducible cytochrome P450 enzyme activity. |

| Applications |
1. A "gold standard" non-tumorigenic in vitro model for studying hepatic lipid metabolism, fatty liver disease (NAFLD/MASLD), and lipotoxicity; 2. Screening for drug-induced liver injury (DILI), hepatotoxicity, and PPARα agonists; 3. Elucidating mechanisms of insulin signaling pathways, hepatic gluconeogenesis, glycogen synthesis, and glucose output; 4. Investigating drug biotransformation, cytochrome P450 (CYP450) induction, and Phase II detoxification enzymes; 5. Establishing models for acute hepatic stress, ischemia-reperfusion injury, and the dynamic processes of liver regeneration. |

| Characteristics |
| Tumorigenic |
No, non-tumorigenic under standard physiological baselines; displays contact inhibition and typical non-malignant hepatocyte morphology. |
| Transformant |
Human transforming growth factor-alpha (TGF-α) driven by the mouse albumin promoter (Alb-1). |
| Functional Hepatocyte Markers |
Endogenously expresses key liver-specific markers and serum proteins, including albumin, α1-antitrypsin, transferrin, and cytokeratin 18 (CK18). |
| Ultrastructure & Metabolism |
Electron microscopy confirms rich, functional intracellular organelles typical of primary hepatocytes: abundant peroxisomes, rough and smooth endoplasmic reticulum, mitochondria, and glycogen storage granules. |
| Growth Kinetics |
Moderate to rapid adherent expansion; population doubling time spans approximately 24 to 36 hours. |

| Culture Conditions and Handling |
| Subculturing |
1. Completely aspirate and discard the complete culture medium from the culture vessel. 2. Gently rinse the cell monolayer with sterile, Ca2+/Mg2+-free PBS to remove residual serum containing trypsin inhibitors. 3. Add 1.0 to 2.5 mL of 0.25% trypsin-0.53 mM (0.02%) EDTA solution, pre-warmed to the appropriate temperature, ensuring the cell layer is completely covered. 4. Incubate at 37°C for 3 to 7 minutes; monitor under an inverted microscope until the cells round up, loosen, and completely detach. 5. Add an equal volume of pre-warmed complete growth medium to immediately neutralize trypsin activity. 6. Gently pipette the cell suspension to disperse the cells, centrifuge at approximately 125 × g for 5 minutes, carefully discard the supernatant, gently resuspend the cells in fresh complete growth medium, and distribute them into new culture vessels. |
| Thawing Protocol Note |
AML-12 cells are sensitive to pH changes during early post-thaw recovery. Prior to introducing thawed vial contents, place the culture vessel containing fresh complete growth medium into the 5% CO2 incubator for at least 15 minutes to allow the medium to equilibrate to its physiological pH range (7.0 to 7.6). |

| Subcultivation Ratio |
Split confluent cultures (80% density) at a standard ratio of 1:3 to 1:6. Maintain active growth by subculturing before cells become completely overgrown or piled up. |
