Cell Line Description The GH3 cell line is a rat pituitary tumor cell clone isolated in 1965 from MtT/W5 prolactinomas in female Wistar-Furth rats. It is a well-known model in endocrinology due to its dual ability to synthesize and secrete both growth hormone (GH) and prolactin (PRL). GH3 cells are highly sensitive to a variety of physiological stimuli, including thyrotropin-releasing hormone (TRH), which significantly enhances prolactin secretion and inhibits growth hormone release. Because of these endocrine functional properties, this cell line is widely used to study hormone regulation, signal transduction, and calcium-dependent exocytosis mechanisms.

Culture Conditions and Handling
Subculturing 1. Remove and discard the culture medium.
2. Briefly rinse the cell layer with Ca2+/Mg2+-free PBS or 0.25% trypsin-EDTA solution to remove residual serum.
3. Add trypsin-EDTA solution and observe until the cell layer disperses (usually 5 to 10 minutes).
4. Add complete culture medium and gently pipette to disperse cell clumps.
5. Aliquot the cell suspension into new culture dishes.
Medium Renewal 2 to 3 times per week
Subcultivation Ratio A split ratio of 1:2 to 1:4 is recommended
Culture Medium F-10 Ham's Medium or DMEM/F12 supplemented with 15% Horse Serum and 2.5% Fetal Bovine Serum (FBS).
Culture Conditions Atmosphere: Air, 95%; CO2, 5%; Temperature: 37°C
Cryopreservation Complete growth medium supplemented with 5% to 10% (v/v) DMSO
