HEL92.1.7 Cell Line

General Information
Organism Homo sapiens, human
Cell Line Description HEL 92.1.7 is a human erythroleukemia cell line derived from the peripheral blood of a 30-year-old male patient with erythroleukemia. The cells exhibit a lymphoblast-like or proerythroblast-like morphology and grow in suspension culture either as single cells or in loose clusters. A notable genetic feature of HEL 92.1.7 is the presence of a homozygous JAK2 V617F mutation, making it a classic model for studying the JAK/STAT signaling pathway. Furthermore, this cell line possesses multipotent hematopoietic differentiation potential; upon treatment with specific chemical inducers, HEL 92.1.7 cells can be prompted to synthesize hemoglobin or express megakaryocytic markers. Given its well-defined genomic background and unique hematopoietic phenotype, HEL 92.1.7 is widely used to investigate erythroid and megakaryocytic differentiation pathways, explore the pathogenesis of myeloproliferative neoplasms (MPNs), evaluate JAK2-targeted inhibitors, and conduct high-throughput drug screening.

Culture Conditions and Handling
Subculturing 1. Maintain the cultures by adding fresh medium or replacing the existing medium.
2. Establish the culture system by centrifuging the cells (1,000 rpm, 5 minutes) and resuspending them to achieve a viable cell density of 1 × 10⁵ to 3 × 10⁵ cells/mL.
3. The cell concentration must not exceed 1.5 × 10⁶ cells/mL.
4. Before subculturing (transferring to a new vessel), gently resuspend the cells to prepare a single-cell suspension.
Medium Renewal Every 2 to 3 days
Subcultivation Ratio Maintain cell density between 1 × 10⁵ and 1.5 × 10⁶ cells/mL (split ratio approximately 1:3 to 1:6)
Culture Conditions Atmosphere: Air, 95%; CO2, 5%; Temperature: 37°C
Cryopreservation Complete growth medium supplemented with 5% to 10% (v/v) DMSO
