Cell Line DescriptionThe WEHI-3 cell line is a mouse leukemia cell line specifically derived from the BALB/c strain. It was originally established from a spontaneous myelomonocytic leukemia in mice. The myelomonocytic cell line WEHI-3 constitutively produces a factor that affects the growth and differentiation of cultured mouse B cells. The cell line also secretes colony stimulating factor (CSF), interleukin 1 (IL 1), but not interleukin 2. In a laboratory setting, WEHI-3 cells have been used to evaluate the differentiation potential of various compounds and bioactivity that modulates the hematopoietic system. These cells have proven to be useful in understanding how alterations in gene expression affect myeloid cells and are an important tool in developing therapeutic strategies against myeloid leukemias. The cell line is also used to establish mouse disease models in vivo by transplantation into susceptible mouse strains to study tumor progression and the efficacy of anticancer drugs.

Additional Info4 ng/mL LPS inhibited the growth of WEHI-3, and higher concentrations of LPS blocked its growth.
30 to 40 µg/mL of dextran sulfate also inhibited its growth.
These WEHI-3 cells showed only weak effector activity in antibody-dependent cell-mediated cytotoxicity.

Culture Conditions and Handling
SubculturingCultures can be maintained by adding fresh medium or changing the medium. Alternatively, cultures can be established by centrifugation and resuspending at 2 X 105 viable cells/mL.
Maintain cell density between 2 X 105 and 2 X 106 viable cells/mL. Adherent cells can be harvested by scraping.
