SU-DHL-2 Cell Line

Cell Line DescriptionSU-DHL-2 is a histiocyte cell line that was isolated in 1974 from the lymph nodes of a 73-year-old Caucasian female with large cell lymphoma. The female had diffuse histiocytic lymphoma (DHL) and after relapse was treated with chemotherapy with prednisone and chlorambucil. This cell line can be used for immunological studies. The majority of these cells are hyperdiploid with a sharp modal number of 51 chromosomes. Occasionally, cells in the hypertetraploid range can also be seen. SU-DHL-2 cells are positive for nonspecific esterase and acid phosphatase. The cells are negative for Epstein-Barr virus nuclear antigen (EBNA-). SU-DHL-2 cells are surface Ig negative (sIG-). In addition, these cells are reported to be E-rosette negative.

KaryotypeMost of these cells were hyperdiploid, with a mode number of chromosomes up to 51, and occasionally cells in the hypertetraploid range were seen. Tiny marker chromosomes were consistently observed, and there was an increase in chromosome number in groups A, B, C, and F.

Culture Conditions and Handling
SubculturingThe culture can be maintained by adding fresh medium. The recommended inoculum size is 1 x 10^5 to 3 x 10^5 cells/mL. Subculture when the cell concentration is between 7 x 10^5 and 1 x 10^6 cells/mL.
