SHP-77 Cell Line

General Information
OrganismHomo sapiens, human
Cell Line DescriptionSHP-77 epithelial cells were derived from a non-encapsulated primary lung tumor in the apex of the left upper lobe of a 54-year-old Caucasian male in 1977. This cell line has unique characteristics, combining morphological features of the undifferentiated large cell variant of small cell lung cancer (SCLC) with typical biochemical properties of classical SCLC. Notably, SHP-77 cells express neural cell adhesion molecule (NCAM), NHK-1, CD56, and Leu-7 (HNK-1; CD57) on their cell surface, in addition to displaying neuroendocrine markers such as L-DOPA decarboxylase and dense core secretory granules. Electron microscopic ultrastructural analysis revealed the presence of glandular formations and intracytoplasmic lamellar bodies. They are suitable for in vitro nude mouse assays to evaluate the immunoreactivity of cells and sera from lung cancer patients. SHP-77 cells can be used as in vitro targets for 51Cr and 111 release cytotoxicity assays and in vivo nude mouse assays to evaluate the immunoreactivity of cells and sera from lung cancer patients. These cells can be used to assess the immune status of SCLC patients receiving radiation or chemotherapy.

Culture Conditions and Handling
SubculturingThe culture can be maintained by adding or replacing fresh medium. Alternatively, the culture can be established by centrifugation and subsequently resuspended at a concentration of approximately 4-5 x 10^5 viable cells/mL. The cells will grow as floating clusters of cells. Some cells may adhere, but only floating cells will be transferred.
Medium RenewalEvery 3 to 4 days
Culture MediumRPMI 1640 + 2mM Glutamine + 10% Heat Inactivated Foetal Bovine Serum (HI-FBS)
Culture ConditionsAtmosphere: Air, 95%; CO2, 5%; Temperature: 37℃
CryopreservationComplete growth medium supplemented with 5% (v/v) DMSO
