Cell Line Description XWLC-05 (Xuanwei Lung Cancer-05) is a human lung adenocarcinoma cell line derived from a 53-year-old female patient in Xuanwei County, Yunnan Province, China. The Xuanwei region is renowned for its unusually high incidence of lung cancer among non-smoking women—a phenomenon attributed to indoor air pollution resulting from the combustion of "smoky coal." XWLC-05 exhibits an epithelial cell morphology and serves as a specialized model for investigating the unique genomic characteristics and environmental etiology of "Xuanwei-type" lung cancer. This cell line is characterized by the presence of specific somatic mutations and epigenetic alterations associated with polycyclic aromatic hydrocarbons (PAHs). As an indispensable research tool, this cell line plays a pivotal role in fields such as the mechanistic study of non-smoking-related carcinogenesis, the genetics of cancer within specific populations, and the screening of targeted therapeutic agents for distinct subtypes of lung adenocarcinoma.

Culture Conditions and Handling
Subculturing 1. Remove and discard the culture medium.
2. Briefly rinse the cell monolayer with PBS (Ca²⁺/Mg²⁺-free) to thoroughly remove residual serum.
3. Add 2.0 to 3.0 mL of 0.25% Trypsin–0.53 mM EDTA solution, and observe under an inverted microscope until the cell monolayer has dispersed (typically requiring 3 to 8 minutes).
4. Add 6.0 to 8.0 mL of complete growth medium and gently pipette to prepare a single-cell suspension.
5. Aliquot the cell suspension into new culture vessels.
