P3U1 Cell Line

General Information
Organism Mus musculus, mouse
Cell Line Description P3U1 (P3X63Ag8U.1) is a murine myeloma/plasmacytoma cell line derived from a mineral oil-induced MOPC-21 tumor in a BALB/c mouse. Through subclone selection, a variant was obtained that is resistant to 8-azaguanine and deficient in hypoxanthine-guanine phosphoribosyltransferase (HGPRT). P3U1 is one of the most commonly used fusion partner cell lines for the generation of monoclonal antibody-producing hybridomas; because it neither synthesizes nor secretes immunoglobulin chains and cannot survive in HAT (hypoxanthine-aminopterin-thymidine) selection medium, it facilitates the efficient enrichment of hybridomas following cell fusion. The cell line grows in suspension and exhibits a lymphoblast-like morphology. P3U1 is utilized for the construction of stable transfectomas and is widely applied in research areas such as somatic cell hybridization, the engineering of antibody-secreting cells, and the genetics of B-cell myelomas. As a key reference for HGPRT-deficient selection systems and the characterization of hybridoma fusion partners, P3U1 is extensively employed to establish stable transfectomas that express recombinant antibodies or other target proteins.

Culture Conditions and Handling
Subculturing 1. Collect the cell suspension into a centrifuge tube and centrifuge at approximately 200 × g for 5 minutes.
2. Carefully discard the supernatant.
3. Resuspend the cell pellet in fresh complete growth medium and gently pipette to break up any cell aggregates.
4. Count the cells and adjust to the recommended seeding density with fresh medium.
5. Aliquot the cell suspension into new culture vessels.
Medium Renewal 3 times per week
Subcultivation Ratio Maintain at 3–9 × 10⁵ cells/mL; do not exceed 1 × 10⁶ cells/mL. Seed new vessels at approximately 4 × 10⁵ cells/mL.
Culture Conditions Atmosphere: Air, 95%; CO2, 5%; Temperature: 37°C
Cryopreservation Complete growth medium supplemented with 5% (v/v) DMSO
