LA795 Cell Line

General Information
Organism Mus musculus, mouse
Cell Line Description LA-795 is a murine lung adenocarcinoma cell line, established in China in 1979. It originated from a spontaneous papillary lung tumor that developed in a male T739 inbred mouse. Following serial subcutaneous passage in syngeneic T739, 615, and KM mice, the tumor line exhibited a 100% rate of pulmonary metastasis; in 1982, an adherent cell line was isolated from this tumor via primary culture. The cells grow as an adherent monolayer and exhibit an epithelioid, rounded morphology. Their karyotype is hypotetraploid, with a modal chromosome number of approximately 66–69. LA-795 retains the metastatic phenotype of the parental tumor and is tumorigenic in immunocompetent syngeneic mice. Consequently, it is highly valuable for studying spontaneous hematogenous metastasis, tumor-stroma interactions, and for in vivo fluorescence imaging using variants labeled with eGFP (enhanced green fluorescent protein). This cell line serves as a critical syngeneic model for screening anti-angiogenic agents and immunomodulatory compounds, and it is widely used in research concerning lung cancer invasion, chromosomal instability, and the preclinical evaluation of anti-tumor drugs.

Culture Conditions and Handling
Subculturing 1. Remove and discard the culture medium.
2. Briefly rinse the cell monolayer with PBS (Ca²⁺/Mg²⁺-free) to thoroughly remove residual serum.
3. Add 2.0 to 3.0 mL of 0.25% Trypsin–0.53 mM EDTA solution, and observe under an inverted microscope until the cell monolayer has dispersed (typically takes 2 to 5 minutes).
4. Add 6.0 to 8.0 mL of complete growth medium, and gently pipette to prepare a single-cell suspension.
5. Aliquot the cell suspension into new culture vessels.
Medium Renewal At subculturing, or 2 to 3 times per week
Subcultivation Ratio A split ratio of 1:3 is recommended when the monolayer reaches approximately 80% confluence.
Culture Conditions Atmosphere: Air, 95%; CO2, 5%; Temperature: 37°C
Cryopreservation 95% complete growth medium + 5% DMSO
