LS411N Cell Line

General Information
Organism Homo sapiens, human
Cell Line Description LS411N is a human colorectal adenocarcinoma cell line established by L. Suardet in 1985 at the Swiss Institute for Experimental Cancer Research. The cell line was derived from a primary cecal tumor resected from a 32-year-old Caucasian male patient, who was diagnosed with a poorly differentiated, Dukes stage B cecal adenocarcinoma. The cells exhibit a typical epithelial morphology and grow as an adherent monolayer. Molecular characterization of LS411N reveals a homozygous BRAF p.V600E mutation, biallelic loss of TP53 and PTEN, and a truncating mutation in the APC gene, while the KRAS and PIK3CA genes remain wild-type. The cell line displays high microsatellite instability (MSI-H)—a relatively rare feature in BRAF-mutated colorectal tumors—making it an ideal model for studying mismatch repair-deficient (dMMR), BRAF-driven disease. LS411N is tumorigenic in immunodeficient mice and possesses a near-triploid karyotype, with a modal chromosome number of approximately 73–75. This cell line is widely used in research concerning the efficacy of BRAF and MEK inhibitors, the mechanisms of resistance to EGFR-pathway-targeted therapies, and the preclinical evaluation of combination treatment regimens for MSI-H colorectal cancer.

Culture Conditions and Handling
Subculturing 1. Remove and discard the culture medium.
2. Briefly rinse the cell monolayer with PBS (Ca²⁺/Mg²⁺-free) to thoroughly remove residual serum.
3. Add 2.0 to 3.0 mL of 0.25% Trypsin–0.53 mM EDTA solution, and observe under an inverted microscope until the cell monolayer has dispersed (typically takes 2 to 5 minutes).
4. Add 6.0 to 8.0 mL of complete growth medium, and gently pipette to prepare a single-cell suspension.
5. Aliquot the cell suspension into new culture vessels.
Medium Renewal 2 to 3 times per week
Subcultivation Ratio A split ratio of 1:3 is recommended.
Culture Conditions Atmosphere: Air, 95%; CO2, 5%; Temperature: 37°C
Cryopreservation Complete growth medium supplemented with 5% (v/v) DMSO
