Cell Line DescriptionMM.1S is a B lymphoblastoid cell line derived in 1986. The blast cell line, MM.1, was established from the peripheral blood of a multiple myeloma patient who had become resistant to steroid therapy. MM.1S was derived from a 42-year-old African American female and has been documented to express CD25, CD38, CD52, and CD59. It also expresses the glucocorticoid receptor and is sensitive to dexamethasone. The MM.1S cell line provides an important framework for understanding the underlying biology, progression, and therapeutic interventions of this disease. MM.1S exhibits key molecular features and genetic mutations commonly found in multiple myeloma patients and is a representative model for preclinical studies. Its in vitro and in vivo behavior elucidates the proliferation dynamics of myeloma cells, their interactions in the bone marrow microenvironment, and their responses to various therapeutic agents. Researchers have extensively utilized MM.1S to gain insight into the pathogenesis of the disease, investigate the efficacy of novel therapeutic compounds, and elucidate mechanisms of resistance in multiple myeloma.

Culture Conditions and Handling
SubculturingThis is a mixed cell culture. Cells can be grown either as a lightly attached monolayer or in suspension. The culture can be maintained by adding fresh medium. Alternatively, subcultures can be prepared by scraping adherent cells into medium containing floating cells, collecting the cells by centrifugation, resuspending the cell pellet in fresh medium and dispensing into new flasks.
Note: Subculture before cells become confluent. Cells in over-confluent cultures begin to form rosettes with necrotic centers.
Subcultivation RatioThe ratio of 1:2 to 1:4 is recommended.
