Cell Line DescriptionEMT6 is a murine mammary carcinoma cell line derived from transplanted hyperplastic alveolar nodules in BALB/c mice. In syngeneic mice, EMT6 cells form tumors and metastasize spontaneously, primarily to the lungs. EMT6 cells are highly adaptable to various growth conditions and have a relatively high mitotic index, which facilitates easy culture and experimental manipulation in a laboratory environment. EMT6 also offers the advantage of orthotopic implantation. This means the cells can be implanted directly into the mammary fat pads of mice, closely mimicking the natural growth environment of mammary tumors. Orthotopic implantation of EMT6 enables researchers to study tumor progression, invasion, metastasis, and treatment response in a more clinically relevant setting. This model helps evaluate new treatment strategies and treatment efficacy, provides valuable insights into breast cancer development and improves translational research efforts. Due to their apparent sensitivity to radiation, they are also used in radiobiological research. The EMT6 cell line played an important role in the development of hypoxic cell sensitizer protocols and has been used to test the efficacy of photodynamic therapy agents.

Additional InfoRecently, EMT6 has emerged as a valuable preclinical model for immuno-oncology research in triple-negative breast cancer. EMT6 tumors express PD-L1 and are moderately responsive to immunotherapy. Checkpoint inhibitors (anti-CTLA-4 or anti-PD-L1) alone often have little effect on tumor growth, while combination therapies have shown greater success, making EMT6 a useful model for combination therapy studies.

DerivationEMT6 was established from transplantable mouse mammary carcinomas generated in BALB/cCRGL mice following implantation of hyperplastic mammary alveolar nodules. The resulting tumor line was propagated in BALB/cKa mice and adapted to tissue culture after the 25th passage of animals, and the cell line was named EMT. EMT6 is a clonal isolate of EMT isolated in 1971 at Stanford University.

Culture Conditions and Handling
Subculturing1. Remove old medium from adherent cells and wash with calcium- and magnesium-free PBS. For T25 flasks, use 3-5 ml of PBS and for T75 flasks, use 5-10 ml.
2. Then, completely cover the cells with Accutase, using 1-2 ml for T25 flasks and 2.5 ml for T75 flasks.
3. Allow cells to detach by incubating at room temperature for 8-10 minutes.
4. After incubation, cells were gently mixed with 10 ml of medium to resuspend and centrifuged at 300xg for 3 min.
5. Discard the supernatant, resuspend the cells in fresh medium, and transfer them to a new flask that already contains fresh medium.

Subcultivation RatioThe ratio of 1:4 to 1:10 is recommended.
Culture MediumDMEM: Ham's F12, w: 3.1 g/L Glucose, w: 1.6 mM L-Glutamine, w: 15 mM HEPES, w: 1.0 mM Sodium pyruvate, w: 1.2 g/L NaHCO3; To make the complete growth medium, add the following components to the base medium: fetal bovine serum to a final concentration of 10%.
