SNU-C1 Cell Line

General Information
Organism Homo sapiens, human
Cell Line Description SNU-C1 is a human colorectal adenocarcinoma cell line established at Seoul National University in 1984. It was derived from the ascites of a 71-year-old Korean male patient, whose primary tumor was a moderately differentiated adenocarcinoma of the colon. It is one of the earlier members of the SNU series of colorectal cancer cell lines. In culture, these cells grow in suspension, forming round cell clusters. SNU-C1 harbors a homozygous TP53 p.Ser166Ter nonsense mutation and an in-frame APIP–SLC1A2 fusion; it is microsatellite stable (MSS). This cell line is also a widely cited model for studying chromothripsis, characterized by hundreds of clustered chromosomal rearrangements concentrated on chromosome 15. It is extensively used in research areas including gastrointestinal cancer biology, large-scale pharmacogenomics and CRISPR-based dependency screening, molecular subtyping of colorectal cancer, proteogenomic analysis, and cytogenetic studies of large-scale genomic rearrangements.

Culture Conditions and Handling
Subculturing 1. Collect the cell suspension into a centrifuge tube and centrifuge at approximately 200 × g for 5 minutes.
2. Carefully discard the supernatant.
3. Resuspend the cell pellet in fresh complete growth medium and gently pipette to break up any cell aggregates.
4. Count the cells and adjust to the recommended seeding density with fresh medium.
5. Aliquot the cell suspension into new culture vessels.
Medium Renewal Every 3 to 5 days
Subcultivation Ratio Dilute the cell suspension with fresh complete growth medium at a ratio of 1:2 to 1:4; refer to the batch-specific information for the recommended dilution.
Culture Conditions Atmosphere: Air, 95%; CO2, 5%; Temperature: 37°C
Cryopreservation Complete growth medium supplemented with 5% (v/v) DMSO
