NCI-H146 Cell Line

General Information
Organism Homo sapiens, human
Cell Line Description NCI-H146 is a human small-cell lung cancer (SCLC) cell line established in 1979 by A.F. Gazdar and J.D. Minna at the NCI–Navy Medical Oncology Branch. The cell line was derived from a bone marrow metastasis in a 59-year-old white male patient with SCLC. These cells grow in suspension culture as floating multicellular aggregates and are tumorigenic in nude mice. NCI-H146 retains the neuroendocrine features characteristic of SCLC, including high expression of neuron-specific enolase, brain-type creatine kinase, and L-dopa decarboxylase, as well as bombesin-like immunoreactivity; the cells also test positive for keratin and vimentin. Genetically, the cell line harbors a homozygous truncating mutation in RB1, a heterozygous frameshift mutation in TP53, and two truncating mutations in KMT2D; it is classified as microsatellite stable (MSS). NCI-H146 is widely used in SCLC research, including studies on neuropeptide autocrine signaling, BCL-2 family inhibitors, PARP1-targeted therapies, responses to etoposide-platinum combination chemotherapy, and DNA demethylation strategies.

Culture Conditions and Handling
Subculturing 1. Collect the cell suspension into a centrifuge tube and centrifuge at approximately 200 × g for 5 minutes.
2. Carefully discard the supernatant.
3. Resuspend the cell pellet in fresh complete growth medium and gently pipette to break up any cell aggregates.
4. Count the cells and adjust to the recommended seeding density with fresh medium.
5. Aliquot the cell suspension into new culture vessels.
Medium Renewal 2 to 3 times per week
Subcultivation Ratio Maintain at 1-2 × 10⁵ cells/mL; split at a ratio of 1:2 to 1:6 when aggregates become dense.
Culture Conditions Atmosphere: Air, 95%; CO2, 5%; Temperature: 37°C
Cryopreservation Complete growth medium supplemented with 5% (v/v) DMSO
