Cell Line DescriptionCOLO 205 is a human colon cancer cell line originally isolated in 1957 from the ascites of a 70-year-old white male colon cancer patient who had been treated with 5-fluorouracil for several weeks. This cell line is characterized by its epithelial morphology and is frequently used in biomedical research focused on colorectal cancer, particularly related to cancer biology, drug response, and metastasis mechanisms. COLO 205 cells exhibit a hyperdiploid karyotype and are known to form moderately well-differentiated adenocarcinomas when xenografted into immunodeficient mice. COLO 205 cells express several key oncogenic and tumor suppressor pathways, making them a valuable model for pharmacological testing and cancer research. They are sensitive to tumor necrosis factor-related apoptosis-inducing ligand (TRAIL), making them suitable for apoptosis studies. In addition, these cells have been widely used to study the pharmacodynamics of various chemotherapeutic drugs, providing insights into the action and resistance mechanisms of colorectal cancer treatments.

Additional InfoThe COLO 205 cell line is derived from Dukes' type D tumor and expresses a 36,000 dalton cell surface glycoprotein. These cells were negative for CSAp expression and positive for keratin expression. The COLO 205 cell line is used to study SV-40 monkey virus and provides a suitable host for studying colon cancer. COLO 205 cells are tumorigenic in nude mice and were used to create a cell line-derived COLO 205 xenograft mouse model. The COLO 205 colon tumor model is an established model for studying the in vivo efficacy of 5-FU or bevacizumab.

Culture Conditions and Handling
Subculturing1. Briefly rinse the cell layer with 0.25% (w/v) Trypsin-0.53 mM EDTA solution to remove all traces of serum containing trypsin inhibitors.
2. Add 2.0 to 3.0 mL of trypsin-EDTA solution to the flask and observe the cells under an inverted microscope until the cell layer is dispersed (usually within 5 to 10 minutes). NOTE: To avoid clumping, do not agitate the cells by knocking or shaking the flask while waiting for cells to detach. Cells that are difficult to separate can be placed at 37°C to facilitate dispersion.
3. Add 6.0 to 8.0 mL of complete growth medium and aspirate cells by gentle pipetting.
4. Add an appropriate amount of cell suspension to the centrifuge tube containing floating cells. Centrifuge at 125 x g for 5 to 10 minutes. Resuspend in fresh medium and plate at appropriate passage ratio.
5. Incubate cultures at 37°C.
NOTE: The volumes used in this protocol are for a 75 cm2 flask. For other sizes of culture vessels, proportionally reduce or increase the amount of dissociation medium.
