Cell Line DescriptionBeWo is a cell line that exhibits epithelial morphology and was isolated from the placenta of a patient with choriocarcinoma. The b30 subclone of these cells can be grown on permeable membranes in double chambers to form confluent cell layers, allowing the rate of nutrient entry into the cell from the apical surface and efflux from the basement membrane to be determined. BeWo cells secrete a variety of hormones, including human chorionic gonadotropin (hCG), human chorionic growth hormone (placental lactogen), and steroid hormones such as estrone, estriol, and estradiol. However, the levels of β-hCG and estradiol secreted by BeWo cells are lower than those of other choriocarcinoma cell lines, such as JEG-3. Following forskolin treatment, β-hCG secretion in BeWo cells is increased to levels similar to those observed in other choriocarcinoma cell lines. In addition, forskolin treatment also increases the levels of progesterone secreted by BeWo cells. These cells have been widely used as an in vitro model to study the uptake of various nutrients into the placenta, including glucose, amino acids, and iron.

Culture Conditions and Handling
SubculturingThe volumes given are for 75 cm2 flasks. For culture vessels of other sizes, increase or decrease the amount of dissociation medium required proportionally.
1. Remove and discard the culture medium.
2. Briefly rinse the cell layer with 0.25% (w/v) trypsin-0.53 mM EDTA solution to remove all traces of serum containing trypsin inhibitors.
3. Add 2.0 to 3.0 mL of trypsin-EDTA solution to the flask and observe the cells under an inverted microscope until the cell layer is dispersed (usually within 5 to 15 minutes).
Note: To avoid clumping, do not agitate the cells by tapping or shaking the flask while waiting for the cells to detach. Cells that are difficult to detach can be placed at 37℃ to facilitate dispersion.
4. Add 6.0 to 8.0 mL of complete growth medium and gently remove the cells by pipetting.
5. Add an appropriate amount of the cell suspension to a new culture vessel.
6. Incubate the culture at 37℃.
