NCI-H1703 Cell Line

General Information
Organism Homo sapiens, human
Cell Line Description NCI-H1703 is a human lung squamous cell carcinoma cell line established in 1987 by A.F. Gazdar and J.D. Minna at the NCI–Navy Medical Oncology Branch, located at the National Naval Medical Center in Bethesda, Maryland. The cell line was derived from a 54-year-old Caucasian male patient with stage 1 lung cancer and a 50 pack-year smoking history. These cells grow as an adherent epithelial monolayer with a doubling time of approximately 48 hours. The genetic profile of NCI-H1703 includes a homozygous alteration at the splice donor site of TP53 (at the boundary of intron 8) and a homozygous CDKN2A p.Asp84Val mutation—a combination of mutations characteristic of lung squamous cell carcinoma. Furthermore, the cell line is microsatellite stable. It is widely used in research concerning the biology of lung squamous cell carcinoma, p53 mutations, proteomic and secretomic profiling, phosphotyrosine signaling pathways, as well as large-scale pharmacogenomic studies and CRISPR-based genetic dependency screens.

Culture Conditions and Handling
Subculturing 1. Remove and discard the culture medium.
2. Briefly rinse the cell monolayer with PBS (Ca²⁺/Mg²⁺-free) to thoroughly remove residual serum.
3. Add 2.0 to 3.0 mL of 0.25% Trypsin–0.53 mM EDTA solution, and observe under an inverted microscope until the cell monolayer has dispersed (typically takes 2 to 5 minutes).
4. Add 6.0 to 8.0 mL of complete growth medium, and gently pipette to prepare a single-cell suspension.
5. Aliquot the cell suspension into new culture vessels.
Medium Renewal 2 to 3 times per week
Subcultivation Ratio A split ratio of 1:3 to 1:6 is recommended.
Culture Conditions Atmosphere: Air, 95%; CO2, 5%; Temperature: 37°C
Cryopreservation Complete growth medium supplemented with 5% (v/v) DMSO
