SK-LU-1 Cell Line

General Information
Organism Homo sapiens, human
Cell Line Description SK-LU-1 is a human lung adenocarcinoma cell line established at the Memorial Sloan Kettering Cancer Center. It was derived from a primary lung tumor resected from a 60-year-old Caucasian female patient and is characterized as poorly differentiated (grade III). The cells grow as an adherent epithelial monolayer and have been extensively utilized in the National Cancer Institute’s (NCI) drug-screening platform, as well as in pharmacological research on non-small cell lung cancer (NSCLC). Genetically, SK-LU-1 harbors a heterozygous KRAS p.G12D activating mutation, a homozygous TP53 p.H193R hotspot mutation, and an ARID1A frameshift mutation that results in a truncated protein; the EGFR gene remains wild-type. The cell line exhibits the alternative lengthening of telomeres (ALT) mechanism, possesses a subtetraploid karyotype, and contains multiple marker chromosomes. SK-LU-1 is tumorigenic in immunotolerant rats and is classified as microsatellite stable (MSS). It is widely used in research concerning the biology of KRAS-driven NSCLC, high-throughput pharmacological screening, CRISPR-based functional genomic screening, and the assessment of drug responses in in vivo xenograft models.

Culture Conditions and Handling
Subculturing 1. Remove and discard the culture medium.
2. Briefly rinse the cell monolayer with PBS (Ca²⁺/Mg²⁺-free) to thoroughly remove residual serum.
3. Add 2.0 to 3.0 mL of 0.25% Trypsin–0.53 mM EDTA solution, and observe under an inverted microscope until the cell monolayer has dispersed (typically takes 2 to 5 minutes).
4. Add 6.0 to 8.0 mL of complete growth medium, and gently pipette to prepare a single-cell suspension.
5. Aliquot the cell suspension into new culture vessels.
Medium Renewal 2 times per week
Subcultivation Ratio A split ratio of 1:2 to 1:4 is recommended when the monolayer reaches 70–80% confluence.
Culture Conditions Atmosphere: Air, 95%; CO2, 5%; Temperature: 37°C
Cryopreservation Complete growth medium supplemented with 5% (v/v) DMSO
