KYSE-180 Cell Line

General Information
Organism Homo sapiens, human
Cell Line Description KYSE-180 is a human esophageal squamous cell carcinoma (ESCC) cell line established by Yutaka Shimada and colleagues at the First Department of Surgery, Kyoto University Faculty of Medicine. The cell line was derived from a primary tumor located in the mid-thoracic esophagus of a 53-year-old Japanese male patient, who was diagnosed with a well-differentiated ESCC (stage T4N1M0). These cells grow as an adherent monolayer and exhibit an epithelial-like morphology; they have a doubling time of approximately 40 to 50 hours and are tumorigenic in immunodeficient mice. KYSE-180 harbors a homozygous TP53 p.Ile195Thr mutation and exhibits CCND1 (cyclin D1) amplification. The cell line has a hyperdiploid karyotype with a modal chromosome number of 55 and is microsatellite-stable. It is widely used in ESCC research, including studies on radiosensitivity, autocrine nerve growth factor signaling, chromosomal instability and LRP1B, combined radiotherapy and anti-angiogenic therapy, as well as cancer pharmacogenomics and CRISPR-based screening.

Culture Conditions and Handling
Subculturing 1. Remove and discard the culture medium.
2. Briefly rinse the cell monolayer with PBS (Ca²⁺/Mg²⁺-free) to thoroughly remove residual serum.
3. Add 2.0 to 3.0 mL of 0.25% Trypsin–0.53 mM EDTA solution, and observe under an inverted microscope until the cell monolayer has dispersed (typically takes 2 to 5 minutes).
4. Add 6.0 to 8.0 mL of complete growth medium, and gently pipette to prepare a single-cell suspension.
5. Aliquot the cell suspension into new culture vessels.
Medium Renewal Every 3 to 5 days
Subcultivation Ratio Split confluent cultures at a ratio of 1:5 to 1:10; seed at approximately 2–3 × 10⁶ cells per 80 cm².
Culture Conditions Atmosphere: Air, 95%; CO2, 5%; Temperature: 37°C
Cryopreservation 70% complete growth medium + 20% FBS + 10% DMSO
