Cell Line Description SCC-9 is a human squamous cell carcinoma cell line derived from the tongue of a 25-year-old male patient. The cells display an epithelial morphology, grow adherently in culture, and are widely utilized in head and neck cancer research, particularly for studying oral cavity squamous cell carcinoma biology. SCC-9 cells exhibit gene expression of epidermal keratins as well as low levels of involucrin. They harbor genomic alterations, including mutations in the TP53 tumor suppressor gene. The line demonstrates robust tumorigenic capacity, forming tumours within 21 days at high frequency when inoculated subcutaneously into immunocompromised mice. While growth can be enhanced using feeder layers under specific conditions, SCC-9 cells generally do not grow well in semi-solid media. This line serves as a valuable model for investigating oral oncogenesis, evaluating novel chemotherapeutic agents, and conducting target identification through genome-wide screening strategies.

Culture Conditions and Handling
Subculturing 1. Remove and discard the culture medium.
2. Briefly rinse the cell monolayer with PBS (without Ca2+/Mg2+) to completely remove residual serum.
3. Add 1.0 to 2.0 mL of 0.25% trypsin-0.03% EDTA solution and incubate the culture vessel at room temperature or 37°C until the cell monolayer detaches.
4. Add fresh complete medium to neutralize the trypsin and gently pipette up and down to prepare a single-cell suspension.
5. Subculture the cell suspension into new culture vessels.
