Cell Line DescriptionSK-MEL-5 is a human melanoma cell line derived from a metastatic axillary lymph node of a young female patient. The cell line was established in 1974 from a metastatic site (axillary lymph node) of malignant melanoma in a 24-year-old Caucasian female. It is characterized by the V600E mutation of BRAF and high expression of wild-type NRAS, as well as relatively high levels of ABCB1 transcripts. SK-MEL-5 cells have been used in various studies to explore new therapies against melanoma and have been used in various in vitro experiments.

Culture Conditions and Handling
Subculturing1. Remove old medium from adhered cells and wash with PBS without calcium and magnesium. For T25 flasks, use 3-5 ml PBS and for T75 flasks, use 5-10 ml.
2. Then completely cover the cells with Accutase, using 1-2 ml for T25 flasks and 2.5 ml for T75 flasks.
3. Allow cells to incubate at room temperature for 8-10 minutes to allow them to detach.
4. After incubation, gently mix the cells with 10 ml medium to resuspend, then centrifuge at 300 x g for 3 minutes.
5. Discard the supernatant, resuspend the cells with fresh medium, and transfer them to a new flask already containing fresh medium.
Medium Renewal2 to 3 times per week
Subcultivation RatioThe ratio of 1:3 to 1:6 is recommended.
Culture MediumEMEM, w: 2 mM L-Glutamine, w: 1.5 g/L NaHCO3, w: EBSS, w: 1 mM Sodium pyruvate, w: NEAA. To make the complete growth medium, add the following components to the base medium: fetal bovine serum to a final concentration of 10%.
