BT474 Cell Line

Cell Line Description BT-474 is a human breast cancer cell line established in 1978 by E.Y. Lasfargues and his colleagues. This cell line was isolated from the solid primary invasive ductal carcinoma tissue of a 60-year-old Caucasian female patient. BT-474 cells grow as an adherent monolayer, typically forming dense, multilayered epithelial patches or colonies, and rarely achieve a completely flat, confluent state. Internationally, this cell line is recognized as the preferred foundational in vitro model for HER2-positive (Luminal B subtype) breast cancer. Given its simultaneous high expression of hormone receptors coupled with amplification of the Human Epidermal Growth Factor Receptor (HER2), BT-474 has emerged as a pivotal preclinical targeted cell line model, frequently utilized to evaluate targeted therapies—such as Trastuzumab and Selective Estrogen Receptor Modulators (SERMs)—as well as to screen for novel small-molecule kinase inhibitors.

Tumorigenic Yes, forms progressively growing tumors when engrafted into immunocompromised mice or rats. Note: In vivo growth is often enhanced by estrogen supplementation.

Culture Conditions and Handling
Subculturing 1. Completely aspirate and discard the spent complete culture medium.
2. Gently rinse the cell monolayer with a standard 0.25% Trypsin–0.53 mM EDTA solution to remove residual traces of serum containing active trypsin inhibitors.
3. Aspirate the rinse solution, add 1.0 to 2.0 mL of fresh Trypsin–EDTA solution to completely immerse the cell layer, and allow it to stand undisturbed.
4. Incubate at room temperature or 37°C until the cells round up and detach. Critical Tip: BT-474 cells are highly prone to physical aggregation. While waiting for the cells to detach, never agitate the cells by tapping, vibrating, or shaking the culture flask, as this will result in irreversible cell clumping.
5. Add 6.0 to 8.0 mL of complete growth medium to the culture vessel, and gently resuspend the cells by repeated pipetting to ensure uniform dispersion.
6. Centrifuge the cell suspension at approximately 125 × g for 5 to 7 minutes; carefully aspirate the supernatant, gently resuspend the cell pellet in fresh medium, and then aliquot the suspension into new culture vessels.

Subcultivation Ratio A low subcultivation ratio of 1:2 to 1:3 is standardly recommended due to its slow, clustered growth kinetics.
