NB4 Cell Line

General Information
Organism Homo sapiens, human
Cell Line Description NB4 is a human acute promyelocytic leukemia (APL; AML FAB M3 subtype) cell line, established in 1989 from a bone marrow sample taken from a 23-year-old Caucasian female patient during her second relapse. This cell line serves as a reference model for studying the PML–RARA fusion oncoprotein, which arises from the t(15;17)(q22;q11–12) translocation. NB4 cells grow in suspension and exhibit a round, pleomorphic morphology; upon treatment with all-trans retinoic acid (ATRA), they undergo terminal differentiation into granulocytes—a process that forms the molecular basis for differentiation therapy in APL. Their immunophenotype is characteristic of immature myeloid cells, expressing CD13, CD33, CD11b, CD4, and CD5, while lacking the expression of CD34 and HLA-DR. ATRA-resistant sublines derived from NB4 (such as NB4-LR1 and NB4-AsR) are essential tools for investigating defects in retinoic acid signaling. NB4 cells are widely used in research concerning ATRA- and arsenic-induced differentiation, resistance to retinoic acid, and targeted therapies directed against the PML–RARA fusion protein.

Culture Conditions and Handling
Subculturing 1. Collect the cell suspension into a centrifuge tube and centrifuge at approximately 200 × g for 5 minutes.
2. Carefully discard the supernatant.
3. Resuspend the cell pellet in fresh complete growth medium and gently pipette to break up any cell aggregates.
4. Count the cells and adjust to the recommended seeding density with fresh medium.
5. Aliquot the cell suspension into new culture vessels.
Medium Renewal 2 to 3 times per week
Subcultivation Ratio Maintain at 0.5-1.0 × 10⁶ cells/mL; split at a ratio of 1:2 to 1:3 every 2 to 3 days.
Culture Conditions Atmosphere: Air, 95%; CO2, 5%; Temperature: 37°C
Cryopreservation 70% complete growth medium + 20% FBS + 10% DMSO
