AU-565 Cell Line

General Information
Organism Homo sapiens, human
Cell Line Description AU-565 is a human breast adenocarcinoma cell line established in the mid-1980s at the Naval Biosciences Laboratory. The cell line was derived from a malignant pleural effusion in a 43-year-old Caucasian female patient who had previously undergone treatment with radiation therapy, corticosteroids, cyclophosphamide, and 5-fluorouracil. AU-565 and its parallel cell line, SK-BR-3—both originating from the same patient—together constitute one of the most widely cited models for HER2-positive breast cancer. These cells grow as an adherent epithelial monolayer with a doubling time of approximately 46 hours. Classified as the "HER2-enriched" subtype, AU-565 exhibits high-level amplification and overexpression of the HER2/ERBB2 gene; the cells test strongly positive for HER2, HER3, HER4, and p53, while testing negative for estrogen and progesterone receptors. The cell line harbors a homozygous TP53 p.R175H hotspot mutation and is microsatellite stable. AU-565 and SK-BR-3 are frequently used as a paired model for HER2-amplified breast cancer in three-dimensional spheroid culture experiments and in vivo xenograft studies to evaluate HER2-targeted therapies. Furthermore, they are commonly included in breast cancer gene expression profiling studies and CRISPR-based screens for target prioritization.

Culture Conditions and Handling
Subculturing 1. Remove and discard the culture medium.
2. Briefly rinse the cell monolayer with PBS (Ca²⁺/Mg²⁺-free) to thoroughly remove residual serum.
3. Add 2.0 to 3.0 mL of 0.25% Trypsin–0.53 mM EDTA solution, and observe under an inverted microscope until the cell monolayer has dispersed (typically takes 2 to 5 minutes).
4. Add 6.0 to 8.0 mL of complete growth medium, and gently pipette to prepare a single-cell suspension.
5. Aliquot the cell suspension into new culture vessels.
Medium Renewal 2 to 3 times per week
Subcultivation Ratio A split ratio of 1:2 to 1:4 is recommended when the monolayer reaches 70–80% confluence.
Culture Conditions Atmosphere: Air, 95%; CO2, 5%; Temperature: 37°C
Cryopreservation Complete growth medium supplemented with 5% (v/v) DMSO
