Cell Line Description HCT-8 (also known as HRT-18) is an epithelial cell line isolated in 1977 from an ileocecal adenocarcinoma of a 67-year-old Caucasian male. It is the gold standard model for studying the biology of human colon cancer, particularly favored for its highly differentiated nature and ability to form tightly connected polarized monolayers of cells. HCT-8 cells are highly sensitive to a variety of chemotherapeutic agents and are often used to study drug resistance mechanisms. Notably, this cell line is also a susceptible host for many human viruses, including coronaviruses such as HCoV-OC43, making it a dual-purpose tool for both oncology and virology research.

Culture Conditions and Handling
Subculturing 1. Remove and discard the culture medium.
2. Quickly wash the cell layer with 0.25% (w/v) trypsin-0.53 mM EDTA solution to remove all serum residue.
3. Add 2.0 to 3.0 mL of trypsin-EDTA solution to the culture flask and observe under an inverted microscope until the cell layer disperses (usually within 5 to 10 minutes).
4. Add 6.0 to 8.0 mL of complete culture medium and gently aspirate the cells using a pipette.
5. Transfer an appropriate amount of cell suspension to a new culture vessel.
Medium Renewal 2 to 3 times per week
Subcultivation Ratio A split ratio of 1:4 to 1:10 is recommended
Culture Medium RPMI 1640 supplemented with 10% Fetal Bovine Serum (FBS), 1 mM sodium pyruvate, and 2 mM L-glutamine.
