Cell Line Description A2780 is one of the most commonly used human ovarian cancer cell lines in biomedical research. It is derived from tumor tissue from untreated ovarian cancer patients. This cell line has an epithelial cell morphology and grows as an adherent monolayer in culture. The A2780 cell line is highly valued for its high sensitivity to various chemotherapeutic drugs, especially cisplatin and carboplatin. Over time, several chemotherapeutic resistant sublineages (such as A2780-CP and A2780-ADR) have been derived from this parent cell line, making it a cornerstone model for studying ovarian tumor drug resistance mechanisms, DNA repair, and the development of novel targeted therapies.

Culture Conditions and Handling
Subculturing 1. Remove and discard the culture medium.
2. Quickly wash the cell layer with Ca2+/Mg2+-free PBS buffer to remove residual serum.
3. Add 2.0 to 3.0 mL of 0.25% trypsin-0.53 mM EDTA solution and observe under a microscope until the cell layer disperses (usually 5 to 10 minutes).
4. Add 6.0 to 8.0 mL of complete culture medium and gently pipette until the cell suspension is a single-cell suspension.
5. Aliquot the cell suspension into new culture containers.

Subcultivation Ratio A split ratio of 1:3 to 1:10 is recommended
Culture Medium RPMI 1640 supplemented with 10% Fetal Bovine Serum (FBS), 2 mM L-glutamine, and 1% Penicillin-Streptomycin.
